Sixty F1 seedlings from a cross between the European pear (Pyrus communis L.) cultivars âOld Homeâ (OH) and âLouise Bonne de Jerseyâ (LBJ) grafted onto Pyrus calleryana and growing in the Plant & Food Research orchard in Motueka, New Zealand (Knäbel et al. 2015), were employed for the detection of polymorphic SNPs for genetic map construction using genotyping by sequencing (GBS).The GBS libraries were prepared using restriction enzyme BamHI, and sequenced on two lanes of Illumina HiSeq2000 platform at Macrogen.
本研究采用的60株杂交F1代实生苗,源自欧洲梨(Pyrus communis L.)品种‘老家园’(Old Home,简称OH)与‘泽西路易丝波娜’(Louise Bonne de Jersey,简称LBJ)的杂交组合,该批材料嫁接至豆梨(Pyrus calleryana)砧木,种植于新西兰莫图伊卡的植物与食品研究所(Plant & Food Research)试验果园中,相关材料信息来自Knäbel等(2015)的研究。本研究通过测序分型(genotyping by sequencing,GBS)技术对该批材料进行多态性单核苷酸多态性(single nucleotide polymorphism,SNP)位点检测,以用于遗传图谱构建;其中GBS文库采用限制性内切酶BamHI进行构建,随后在Macrogen公司的Illumina HiSeq2000测序平台的两个测序泳道上完成上机测序。
