DNA methylation is one of the most studied epigenetic alterations in cancer. Genome-wide DNA methylation profiling was conducted in 6 oral tongue squamous cell carcinomas and matched normal tissues. In the present study, the Illumina Infinium HumanMethylationEPIC BeadChip (EPIC array) was used to characterize the DNA methylation pattern across approximately 850,000 CpG dinucleotide methylation loci using DNA isolated of formalin-fixed and paraffin-embedded tissue sections. Total genomic DNA from formalin-fixed paraffin-embedded (FFPE) tissue sections of oral tongue squamous cell carcinoma (OTSCC) and normal tongue tissue were processed on the Infinium MethylationEPIC array (850K). Samples that passed the quality controls using GenomeStudio and BeadArray Controls Reporter (Illumina), were analyzed to asses their DNA methylation profile.

DNA甲基化是癌症领域研究最为深入的表观遗传改变之一。本研究针对6例舌鳞状细胞癌组织及配对正常组织开展全基因组DNA甲基化谱分析。本次研究采用Illumina Infinium人类甲基化EPIC微珠芯片(Illumina Infinium HumanMethylationEPIC BeadChip,简称EPIC芯片),以从福尔马林固定石蜡包埋组织切片中提取的DNA为材料,对约85万个CpG二核苷酸甲基化位点的DNA甲基化模式进行表征。随后,我们对舌鳞状细胞癌(oral tongue squamous cell carcinoma, OTSCC)及正常舌组织的福尔马林固定石蜡包埋(formalin-fixed and paraffin-embedded, FFPE)组织切片中提取的全基因组DNA,采用Infinium甲基化EPIC芯片(850K芯片)进行上机检测。经GenomeStudio软件与BeadArray阵列质控报告工具(BeadArray Controls Reporter,Illumina)完成质量验证的合格样本,将被用于分析其DNA甲基化谱特征。
