We report the application of single-molecule-based sequencing technology for high-throughput profiling of histone modifications in NSC. By obtaining sequences from chromatin immunoprecipitated DNA, we generated genome-wide chromatin-state maps of NSC. We compared histone modifications between wild type and Sox2-deleted NSC. Overall design: Examination of 2 different histone modifications in 2 cell types.
本研究报道了单分子测序技术(single-molecule-based sequencing technology)在NSC中开展组蛋白修饰(histone modifications)高通量谱分析的应用。通过对染色质免疫沉淀DNA进行测序,我们构建了NSC全基因组范围的染色质状态图谱。本研究对比了野生型(wild type)与Sox2敲除型NSC之间的组蛋白修饰差异。整体实验设计:在2种细胞类型中对2种不同的组蛋白修饰进行检测。
