To investigate the underlying mechanism of pulmonary hypertension, the model of monocrotaline (MCT)-treated pulmonary arterial hypertension (PAH) rats were constructed to detect the differentially expressed profile of genes in lung tissue of PAH rat. The male rats were treated with monocrotaline (MCT) to induce pulmonary arterial hypertension model. One dose of MCT (60 mg/kg) was injected Intraperitoneally into animals. Control rats were injected with the equivalent volume of dissolvent solution according to their weights. After 4 weeks late, the lung tissue was collected to detect the genes expression profile.

为探究肺动脉高压的潜在发病机制,本研究构建了野百合碱(monocrotaline, MCT)诱导的肺动脉高压(pulmonary arterial hypertension, PAH)大鼠模型,以检测PAH大鼠肺组织的基因差异表达谱。实验选取雄性大鼠,按体重单次腹腔注射60 mg/kg剂量的野百合碱以诱导肺动脉高压模型;对照组大鼠则按体重注射等体积的溶媒。造模4周后,采集肺组织以检测基因表达谱。
