Differences of metabolism-related gene expression profiles in human pluripotent stem cells (hPSCs) and hPSC-derived cardiomyocytes (hPSC-CMs) were analyzed and successfully identified. Overall design: 253G4, 201B7, H9, NKX2-5eGFP/w cell lines were used for this experiments. hPSC-CM induction protocol was based on the modulation of the WNT signaling pathway. From Day 13 to Day 17, the cells were incubated with glucose- and glutamine-free medium supplemented with lactate for metabolic selection. hPSC-CMs obtained on Day 30 were used in the experiments.

本研究对人多能干细胞(human pluripotent stem cells, hPSCs)及其诱导分化的心肌细胞(hPSC-derived cardiomyocytes, hPSC-CMs)的代谢相关基因表达谱差异进行了分析并成功完成鉴定。实验总体设计:本实验采用253G4、201B7、H9、NKX2-5eGFP/w共4种细胞系。人多能干细胞向心肌细胞的诱导分化方案基于WNT信号通路的调控构建。在第13天至第17天期间,细胞使用添加乳酸的无糖无谷氨酰胺培养基进行代谢筛选培养。本实验最终采用第30天获取的人多能干细胞衍生心肌细胞开展相关实验。
