The first-generation (F1) hybrid offspring were produced using Scylla paramamosain as the male parent and Scylla serrata as the female parent. Samples of hybrid crabs with displaced eyestalk (DH) and normal eyestalk (NH) were collected for Whole transcriptome resequencing at the first stage of crablet. Finally, a total of six Illumina libraries (3 samples from DH and 3 samples from NH) were constructed and sequenced on the Illumina Hiseq 2500 platform for generating 150bp paired-end reads.
本研究以拟穴青蟹(Scylla paramamosain)为父本、锯缘青蟹(Scylla serrata)为母本,培育获得第一代(F1)杂交后代。采集眼柄异位(displaced eyestalk, DH)与正常眼柄(normal eyestalk, NH)的杂交青蟹样本,于仔蟹早期阶段开展全转录组重测序。最终共构建6个Illumina文库(DH组与NH组各3个样本),在Illumina HiSeq 2500测序平台上完成测序,得到150 bp双端测序读段。
