This study defines the impact of in vivo thrombospondin-1 (THBS1) ablation on diaphram stromal cell profile, histology and contractile funciton in a mosue model of diet-induced obesity Overall design: Single cell RNA sequencing of mononuclear cell isolates from daiphragms of wild type C57 mice fed either control diet or high fat diet (45% kcal from lipid) for 6 months; as well as thrombospondin-1 knockout mice fed high fat diet for 6 months
本研究阐明了体内血小板反应蛋白-1(thrombospondin-1,THBS1)基因敲除对饮食诱导肥胖小鼠模型中膈肌基质细胞谱、组织学特征及收缩功能的影响。整体实验设计:对喂食对照饮食或高脂饮食(热量中45%来自脂质)6个月的野生型C57小鼠膈肌分离得到的单核细胞进行单细胞RNA测序;同时对喂食高脂饮食6个月的血小板反应蛋白-1敲除小鼠开展相同检测。
