Effects of EPA/DHA enriched high fat diets (HFD) compared to HFD-Corn oil after 8 weeks intervention on age/obesity induced skeletal muscle degradation. Results provides novel information on the signalling pathways regulated by EPA and DHA enriched HFD in delaying the onset of muscle degradation in C57Bl/6J mice compared with HFD-Corn oil. Overall design: Four mice representing each diet group on the basis of body weight, plasma triglyceride and plasma cholesterol levels were selected for RNA isolation and microarray analysis. Total RNA from skeletal muscle of these selected animals was purified using the RNeasy® Plus Universal Mini kit (Qiagen Nordic, Sollentuna, Sweden) by following the manufacturer?s instruction carefully. The extracted RNA was quantified by NanoDrop 2000c UV-Vis Spectrophotometer (Thermo Scientific, USA) and the quality of the RNA was estimated by RNA 6000 Nano LabChip for Agilent 2100 Bioanalyzer (Agilent Technologies, Santa Clara, California, USA).

实验设计:依据体重、血浆甘油三酯及血浆胆固醇水平,从每个饮食组中选取4只小鼠用于RNA提取与微阵列分析。严格遵循制造商操作说明书,使用RNeasy® Plus Universal Mini试剂盒(Qiagen Nordic,瑞典索伦图纳)从上述选定小鼠的骨骼肌中纯化总RNA。采用NanoDrop 2000c紫外-可见分光光度计(赛默飞世尔科技,美国)对提取的总RNA进行浓度定量,并使用适配Agilent 2100生物分析仪的RNA 6000 Nano LabChip(安捷伦科技,美国加利福尼亚州圣克拉拉)评估RNA质量。
