To determine the effect of adherence on the metabolic and functional response of human monocytes. Monocytes were stimulated with lipopolysaccharide (LPS) under non-adherent and adherent conditions. To determine the role of glycolysis in LPS-induced immune responses, this pathway was inhibited by glucose deprivation or the glucose analog 2-deoxy-D-glucose (2DG). Monocytes were isolated from 6 healthy volunteers. Each donor's monocytes were exposed to every combination of the three treatment conditions (2 adherene conditions, 2 LPS conditions, 3 glucose conditions).
本研究旨在探究黏附状态对人单核细胞(human monocytes)代谢与功能应答的影响。实验中,单核细胞分别在非黏附与黏附两种条件下经脂多糖(lipopolysaccharide, LPS)刺激。为明确糖酵解在LPS诱导的免疫应答中的作用,本实验通过葡萄糖剥夺或葡萄糖类似物2-脱氧-D-葡萄糖(2-deoxy-D-glucose, 2DG)抑制该通路活性。单核细胞取自6名健康志愿者,每名供者的单核细胞均接受三种处理条件的全部组合:2种黏附条件、2种LPS处理条件及3种葡萄糖处理条件。
