We isolated macrophages from murine lung tumors and their in vitro derived progenitors for RNA sequencing Overall design: Lewis lung carcinoma tumors grown in mice were used to isolate tissue resident CD45+CD11b+Gr1-F4/80hi and bone marrow derived CD45+CD11b+Gr1+F4/80lo macrophages for RNA sequencing. These cells were also used to generate macrophage progenitors by culture in methycellulose + stem cell growth factors. Resulting Colony cells were then sorted for Kit expression and sequenced.
本研究从小鼠肺肿瘤及其体外来源的前体细胞中分离巨噬细胞以开展RNA测序(RNA sequencing)。
实验整体设计如下:以小鼠体内生长的Lewis肺癌(Lewis lung carcinoma)肿瘤为实验材料,分离组织驻留型CD45+CD11b+Gr1-F4/80hi巨噬细胞与骨髓来源型CD45+CD11b+Gr1+F4/80lo巨噬细胞用于RNA测序;同时利用上述细胞在甲基纤维素(methycellulose)联合干细胞生长因子的培养基中培养以制备巨噬细胞前体细胞,随后对形成的集落细胞按Kit表达水平进行分选并完成测序。
