研究目的:对体外进行肝向分化的人多能干细胞(hPSC)开展RNA测序分析,以验证其在分化不同阶段(D0至D21)的分化是否正常。
实验整体设计:将诱导分化得到的肝细胞进行丙型肝炎病毒(HCV)感染实验,以未感染的NINF对照作为参照,并设置辅以各类抗病毒治疗(TEL)或JAK/STAT通路抑制剂治疗(JSI)的组别,随后对样本进行全局基因表达变化分析。本研究重点聚焦于先天免疫基因与干扰素调控基因(IRG)。
所有样本均单独进行测序上机。本实验的研究对象为体外诱导肝向分化的人多能干细胞,分化所得肝细胞接受HCV感染(以未感染的NINF对照为参照),并可辅以或不辅以各类抗病毒治疗(TEL)或JAK/STAT通路抑制剂治疗(JSI)。
