The phosphorylation and dephosphorylation of transcription machinery are essential for the precise control of gene expression. A non-canonical protein phosphatase 2A (PP2A) holoenzyme (denoted INTAC), in which the 14-subunit Integrator recruits RNA polymerase II (Pol II) and the PP2A core enzyme dephosphorylates the C-terminal repeat domain (CTD) of Pol II at Serine-5 and Serine-2. Overall design: We have finished the ChIP-seq and nascent-seq to investigate the role of integrated phosphatase and endonuclease activities in this Integrator-PP2A complex and its function in regulating Pol II phosphorylation and transcription.
转录机器的磷酸化与去磷酸化过程,是实现基因表达精准调控的关键环节。一种非经典的蛋白磷酸酶2A(protein phosphatase 2A, PP2A)全酶(命名为INTAC),由14亚基的整合子(Integrator)招募RNA聚合酶II(RNA polymerase II, Pol II),并由PP2A核心酶在丝氨酸5位(Serine-5)与丝氨酸2位(Serine-2)位点,对Pol II的C端重复结构域(C-terminal repeat domain, CTD)进行去磷酸化修饰。实验整体设计:本研究已完成染色质免疫共沉淀测序(ChIP-seq)与新生转录组测序(nascent-seq),以探究该整合子-PP2A复合体中整合的磷酸酶与核酸内切酶活性的作用,及其在调控Pol II磷酸化与转录过程中的功能。
