We report the different enteroendocrine populations in the large intestine in mice based on their transcriptomics profile. Hormone, receptor and transcription factors are analysed for differential expression between the different cell clusters to give a comprehensive view of the different characterisitcs of the distal entereondocrine cells Overall design: Large intestine epithelium from a Neurod1-crexEYFP mouse was digested into single cells and sorted based on EYFP fluorescence to collect only the enteroendocrine cells. 3500 cells were collected and their transcriptome sequenced using the Chromium system (10x Genomics) and a Hiseq4000 sequencer. Analysis of their transcriptome was perfomed using the cellranger pipeline

本研究基于转录组图谱(transcriptomics profile),报道了小鼠大肠内不同的肠内分泌细胞(enteroendocrine cell)群。研究针对不同细胞簇间的激素、受体及转录因子的差异表达展开分析,以全面解析远端肠内分泌细胞的各项特征。 总体实验设计:取Neurod1-cre×EYFP小鼠的大肠上皮组织,将其消化为单细胞悬液后,基于EYFP荧光进行分选,仅收集肠内分泌细胞。最终收集3500个细胞,采用10x Genomics Chromium单细胞系统与Illumina HiSeq4000测序仪进行转录组测序。转录组数据分析采用Cell Ranger分析流程。
