Capsule switch experiments. Streptococcus pneumoniae

Two pneumococcal isolates had their capsule deleted using Phun-Sweet-Janus and replaced by donor capsules using transformation with gDNA. The isolates were then characterized in growth experiments. From genomic data, the goal is to evaluate additional genes that were exchanged with the capsule, to determine the location of the breakpoints and factors associated with the length of recombination events.

研究人员利用Phun-Sweet-Janus技术敲除两株肺炎链球菌分离株的荚膜，并通过基因组DNA（gDNA）转化的方式，以供体荚膜序列替换原有的敲除位点。随后通过生长实验对改造后的分离株进行特征表征。基于基因组测序数据，本研究的目标为评估与荚膜一同发生重组交换的额外基因，确定重组断点的位置以及与重组事件长度相关的影响因素。
