Zfp296 KO profoundly affects the gene expression patterns of ESCs. ZFP296 belongs to the C2H2-type ZF family. To map the ZFP296 binding sites across the whole genome, chromatin immunoprecipitation DNA-sequencing (ChIP-seq) was performed. Overall design: ChIP-seq for Ty1-tagged ZFP296 expressed in Zfp296-KO #98 ESCs.
Zfp296基因敲除(Zfp296 knockout, KO)可显著影响胚胎干细胞(Embryonic Stem Cells, ESCs)的基因表达谱。ZFP296隶属于C2H2型锌指蛋白家族(C2H2-type Zinc Finger family)。为在全基因组范围内定位ZFP296的结合位点,本研究开展了染色质免疫共沉淀测序(chromatin immunoprecipitation DNA-sequencing, ChIP-seq)实验。实验设计概述:针对在Zfp296基因敲除的#98株胚胎干细胞中表达的、经Ty1标签标记的ZFP296进行ChIP-seq检测。
