WT (C57BL/6) and Ch25h-/- mice were infected i.v. with 1e5 CFU of Listeria monocytogenes. At day 3 post-infection, ~100,000 resident splenic macrophages (CD11b+, Ly6G-, Ly6clow) were isolated by flow cytometery and analyzed by RNA-seq Overall design: Comparative gene expression profiling by RNA-seq of splenic macrophages from C57BL/6 and Ch25h-/- mice infected with Listeria monocytogenes.

野生型（Wild Type，WT）C57BL/6及Ch25h基因敲除（Ch25h knockout，Ch25h-/-）小鼠经静脉（intravenous，i.v.）注射1×10^5菌落形成单位（Colony-Forming Unit，CFU）的单核细胞增生李斯特菌（Listeria monocytogenes）。感染后第3天，通过流式细胞术（flow cytometry）分离得到约100,000个定居型脾脏巨噬细胞（表型为CD11b+、Ly6G-、Ly6clow），并采用RNA测序（RNA-seq）对其进行分析。实验整体设计：针对感染单核细胞增生李斯特菌的C57BL/6及Ch25h-/-小鼠的脾脏巨噬细胞，开展RNA测序比较基因表达谱分析。
