WT (C57BL/6) and Ch25h-/- mice were infected i.v. with 1e5 CFU of Listeria monocytogenes. At day 3 post-infection, ~100,000 resident splenic macrophages (CD11b+, Ly6G-, Ly6clow) were isolated by flow cytometery and analyzed by RNA-seq Overall design: Comparative gene expression profiling by RNA-seq of splenic macrophages from C57BL/6 and Ch25h-/- mice infected with Listeria monocytogenes.
野生型(Wild Type,WT)C57BL/6及Ch25h基因敲除(Ch25h knockout,Ch25h-/-)小鼠经静脉(intravenous,i.v.)注射1×10^5菌落形成单位(Colony-Forming Unit,CFU)的单核细胞增生李斯特菌(Listeria monocytogenes)。感染后第3天,通过流式细胞术(flow cytometry)分离得到约100,000个定居型脾脏巨噬细胞(表型为CD11b+、Ly6G-、Ly6clow),并采用RNA测序(RNA-seq)对其进行分析。实验整体设计:针对感染单核细胞增生李斯特菌的C57BL/6及Ch25h-/-小鼠的脾脏巨噬细胞,开展RNA测序比较基因表达谱分析。
