In the study presented hereCD44CD24-CD44H cells and CD44-CD24CD44L cells were sorted from HMLER cells by flow cytometry. CD44L cells were cultured in low adhesion plate with tumorsphere medium for several generations and we got the new subpopulationCD44LS. RNA sequencing profiling of mRNA levels was performed in these three subpopulation cells.

本研究中，研究人员通过流式细胞术（flow cytometry）从HMLER细胞中分选出CD44CD24⁻CD44H细胞与CD44⁻CD24CD44L细胞。将CD44L细胞接种于低吸附培养板中，采用肿瘤球培养基（tumorsphere medium）进行多代传代培养，最终获得新的细胞亚群CD44LS。随后针对上述三类细胞亚群的信使RNA（messenger RNA）表达水平，开展RNA测序表达谱分析。
