The goal of this experiment was to identify methylation and metabolomic pathways the respond to proton irradiation and examine whether these two pathways coregulate biological pathways. Overall design: DNA was isolated 20 weeks after 0 Gy (sham) or 1 Gy Proton irradiation. 1 ug of hippocamal or left ventricular DNA from 5 mice in each treatment group was pooled, resulting in two pooled biological replicates for each treatment condition. Pooled DNA was subjected to 5hmC DIP and Illumina TrueSeq libraries were constructed.

本实验旨在筛选响应质子辐照的甲基化通路与代谢组通路,并探究二者是否可共同调控生物学通路。 总体实验设计:于接受0 Gy(假照射)或1 Gy质子辐照20周后提取DNA。分别从每个处理组的5只小鼠体内获取1μg海马体或左心室DNA并混合,每个处理条件由此获得两份混合生物学重复。将混合后的DNA进行5-羟甲基胞嘧啶DNA免疫沉淀(5hmC DIP),并构建Illumina TrueSeq文库。
