Analysis of off target generated by CRISPR/Cas9 in DM1 mutant haploid embryonic stem cells

The androgenetic embryonic stem cells we generated carrying several genes mutations could be injected into the oocyte and produce the semi-cloned mice. Those semi-cloned mice could mimic several DM1-associated phenotypes. We try to identify if any off-target exist in those cells inducing by CRISPR/Cas9. Overall design: Examination of off target position in two cell lines.

我们构建的携带多种基因突变的孤雄胚胎干细胞（androgenetic embryonic stem cells）可被注射至卵母细胞中，进而获得半克隆小鼠。该类半克隆小鼠能够模拟多种与DM1相关的表型。本研究旨在探究经CRISPR/Cas9编辑的此类细胞是否存在脱靶位点。整体实验设计：对两种细胞系的脱靶位点开展检测分析。
