This invention relates to the field of biomedicine, and specifically to protein probes and their applications in detecting BACE1 activity. Provided herein are expression vectors comprising genes encoding the following protein fragments: mCherry-N fragment, mCherry-C fragment, VAMP7, and BACE1 recognition peptide. Also provided herein is a fluorescent protein probe that is intuitive, dynamic, simple, highly operable, implementable under physiological conditions, and capable of detecting BACE1 activity in live cells.
