To explore the potential involvement of tRNA derived fragments (tRFs) in the human aortic vascular smooth muscle cells (HASMCs), we conducted tRFs profiling in three pairs of differentiated HASMCs (treated with PBS) and dedifferentiated HASMCs (treated with PDGF-BB 50 ng/ml) by microarray. Our results showed that tRFs were aberrantly expressed in dedifferentiated HASMCs compared with differentiated HASMCs and provided potential targets for novel insights into VSMC differentiation or vascular remodeling diseases. Overall design: The tRFs expression profiles of three paired differentiated HASMCs (treated with PBS) and dedifferentiated HASMCs (treated with PDGF-BB 50 ng/ml)were analyzed using the microarray platform.
为探究tRNA衍生片段(tRNA-derived fragments, tRFs)在人主动脉血管平滑肌细胞(human aortic vascular smooth muscle cells, HASMCs)中的潜在调控作用,我们通过微阵列技术对3组配对的分化型HASMCs(经磷酸盐缓冲液(PBS)处理)与去分化型HASMCs(经50 ng/ml血小板衍生生长因子-BB(PDGF-BB)处理)开展了tRFs表达谱分析。本研究结果显示,相较于分化型HASMCs,去分化型HASMCs中的tRFs表达存在异常,为血管平滑肌细胞分化或血管重构相关疾病的研究提供了潜在靶点与全新研究视角。整体实验设计:采用微阵列平台分析3组配对的分化型HASMCs(经PBS处理)与去分化型HASMCs(经50 ng/ml PDGF-BB处理)的tRFs表达谱。
