Circular RNAs (circRNAs) are an emergent class of non-coding RNAs commonly generated by back-splicing events, resulting in a highly stable RNA structure, devoid of free 5’ and 3’ ends. circRNAs display the main mechanism of action as miRNA sponges, resulting in the modulation of their regulatory activity. Current knowledge suggests that they could play a relevant role in cellular biology and pathophysiology. In this study, for the first time, we detected circRNAs as a new component of the induced pluripotent stem cell (hiPSC)-derived extracellular vesicles (EV) cargo. We further compared the circRNA profile of hiPSC-EV with that of their parental cells (hiPSCs). Two samples: Parental hiPSC versus hiPSC-derived EVs. Parental hiPSC was submitted previously in GSE144629 (reference parental cell).
环状RNA(circRNAs)是一类新兴的非编码RNA,通常由反向剪接事件生成,具备高度稳定的RNA结构,不含游离的5'与3'末端。circRNAs的核心作用机制为充当miRNA海绵,进而调控miRNA的调控活性。现有研究表明,这类RNA在细胞生物学与病理生理学中可能发挥重要作用。
本研究首次检测到环状RNA作为诱导多能干细胞(hiPSC)来源的细胞外囊泡(EV)载荷的新型组分。我们进一步对比了hiPSC来源的细胞外囊泡与其亲本细胞(hiPSCs)的circRNA表达谱。
本研究包含两组样本:亲本hiPSC与hiPSC来源的细胞外囊泡。其中作为参考亲本细胞的亲本hiPSC样本此前已提交至GSE144629数据集。
