Most of our current knowledge about the molecular events ruling plant-virus interaction come from studies focusing on vascular plants. We here characterized the molecular, cellular and physiological events goberning plant-virus interactions in the non-vascular liverwort Marchantia polymorpha. Overall design: 2 weeks old Marchantia plants grown in sterile conditions were incubated with sap from either WT or TMV infected tobacco plants. Two weeks after, samples were collected from infected areas (TMV samples) and anatomically equivalent areas from mock plants (Mock samples). RNA was isolated from 4 pools. mRNA enriched libraries were prepared using Illumina TrueSeq kit and following instructions from manufacturer.

实验设计概述:将生长2周的无菌培养地钱植株,分别用野生型(WT)烟草汁液与烟草花叶病毒(TMV)侵染的烟草汁液进行孵育接种。接种两周后,采集病毒侵染区域的样本(TMV组样本),以及模拟接种组植株对应解剖位置的区域样本(Mock组样本)。从4个混合样本池中提取核糖核酸(RNA),采用Illumina TrueSeq试剂盒并依照厂商说明书构建信使RNA(mRNA)富集文库。
