Arbuscular mycorrhizal (AM) fungal communities were characterized using a meta-barcoding approach. The small subunit ribosomal RNA (SSU rRNA) region was amplified with the barcoded primer pair WANDA/AML2. Amplicons were sequenced using paired-end sequencing (2 × 300 bp) on the Illumina MiSeq platform. Sequence data were processed with the DADA2 denoising algorithm to generate amplicon sequence variants (ASVs). Taxonomic assignment of AM fungal sequences was performed via BLAST searches against the MaarjAM database.

丛枝菌根(Arbuscular Mycorrhizal, AM)真菌群落采用元条形码测序(meta-barcoding)方法进行表征:以带条码的引物对WANDA/AML2扩增小亚基核糖体RNA(SSU rRNA)区域;扩增子序列在Illumina MiSeq测序平台上以2×300 bp双端测序模式进行测序;使用DADA2去噪算法处理测序数据以生成扩增子序列变异体(ASVs);通过针对MaarjAM数据库的BLAST比对完成AM真菌序列的分类学注释。
