We examined the role of HES1 in high salt response as a novel regulator of NFAT5. We analyzed gene expression of HeLa cells under isoosmotic and high salt condition. HeLa cells were transfected with siControl, siNFAT5 and siHES1. mRNA samples were obtained from the transfected cells 7.5 hours after osmotic stress treatment (isoosmotic or high salt). The samples were amplified, labeled and hybridized to the Clariom S Array human (Thermo Fisher Scientific). The data were analyzed with Affymetrix Expression Console Software 1.4.1 offered SST-RMA using Affymetrix default analysis settings. * The data files in the submission were replaced on April 15, 2025 *
本研究考察了HES1作为NFAT5的新型调控因子在高盐应答过程中的作用。我们对等渗与高盐培养条件下的海拉细胞(HeLa)进行了基因表达分析。将海拉细胞分别转染siControl、siNFAT5与siHES1。在渗透压胁迫处理(等渗或高盐)7.5小时后,收集转染细胞的mRNA样本。对样本进行扩增、标记后,与赛默飞世尔科技(Thermo Fisher Scientific)提供的人类Clariom S基因芯片(Clariom S Array human)进行杂交。采用Affymetrix Expression Console Software 1.4.1软件,依托其提供的SST-RMA算法,按照Affymetrix默认分析参数完成数据分析。
