To identify IRF4 transcription factor binding on chromatin at 5' regulatory regions of genes in myeloma cell line models. Keywords: binding site identification design Overall design: Formaldehyde cross-linked, sonicated chromatin is prepared from cell lines Kms12 (test) and Ly19 (control). Chromatin immunoprecipated with anti-IRF4 antibody is labeled with Cy5 and co-hybridized on Agilent Human Promoter Set arrays with chromatin immunoprecipated with normal sera labeled with Cy3 . Two biological replicates were performed for the Kms12 and Ly19 experiments.

实验设计:从细胞系Kms12(实验组)与Ly19(对照组)中制备经甲醛交联、超声破碎的染色质;以抗IRF4抗体进行染色质免疫沉淀得到的产物用Cy5荧光标记,与以正常血清进行免疫沉淀并以Cy3荧光标记的染色质产物共同在安捷伦人类启动子组芯片(Agilent Human Promoter Set arrays)上进行杂交。针对Kms12与Ly19的实验均完成了两次生物学重复。
