crl; Transcriptome comparison of the crl mutant and CDT1-RNAi line

au13-04_cdtbis; cdt1_bis crl mutants and CDT1-RNAi lines have very similar macroscopic phenotypes as well as identical defects in plastid division and biogenesis. Our goal wwas to determine how much of these similarities originated from similar alterations of gene expression. Plantlets of crl mutant and CDT1-RNAi lines were grown in vitro on MS1/2 medium for 14 days. au13-04_cdtbis；cdt1_bis crl突变体与CDT1-RNAi株系在宏观表型上高度相似，且在质体分裂与质体发生过程中存在完全一致的缺陷。本研究旨在明确这些相似性有多少源于基因表达的相似改变。将crl突变体与CDT1-RNAi株系的组培小苗接种于1/2 MS培养基（MS1/2 medium）中培养14天。CDT1-RNAi株系以其对应的野生型（Ws生态型）作为对照，而crl突变体则以其野生型姊妹株为对照，该突变体的遗传背景为Col0生态型。本实验共设置4组染料交换（dye-swap）实验：分别为野生型与RNAi突变体的对照比较，以及野生型与转基因株系的对照比较。
