To investigate the role of CBX3 in the progression of osteosarcoma, we performed a transcriptome analysis of the CBX3-depleted 143B cell line and a matched normal 143B cell line.Differential gene enrichment revealed activation of the anti-DNA damage pathway and activation of the apoptotic and P53 pathways. Overall design: The mRNA profiles of 143B osteosarcoma cells without targeting gene small interfering RNA and targeting CBX3 small molecule interfering RNA were generated by deep sequencing using a Novaseq 6000 sequencer (Illumina).

为探究CBX3在骨肉瘤进展中的作用,我们对CBX3敲低的143B细胞系及匹配的正常143B细胞系开展了转录组分析。差异基因富集分析结果显示,抗DNA损伤通路、细胞凋亡通路及P53通路均被激活。 实验设计概述:采用Illumina Novaseq 6000测序仪对两组143B骨肉瘤细胞进行深度测序,两组分别为转染非靶向小干扰RNA(small interfering RNA)的对照组,以及转染靶向CBX3的小干扰RNA的实验组,以此获取其mRNA表达谱。
