Following total body irradiation, four rhesus macaques were transplanted with lentivirally transduced HSPCs containing unique 35 base-pair barcode sequences in the provirus insertions. Blood was extracted from macaques at time points up through 4 years post-transplant and sorted into cell subsets; targeted DNA sequencing of the barcoded region then allowed for quantification of barcodes in cell subsets.

接受全身照射(total body irradiation)后,四只恒河猴(rhesus macaques)接受了慢病毒转导的造血干细胞及祖细胞(hematopoietic stem and progenitor cells, HSPCs)移植,此类细胞的前病毒整合位点中携带有独特的35碱基对条形码序列(barcode sequences)。研究人员在移植后长达4年的多个时间点采集恒河猴血液,并将血液分选至不同细胞亚群中;随后对带条形码的区域开展靶向DNA测序,即可实现各细胞亚群内条形码的定量检测。
