Elevated COUP-TFII levels are found in Parkinson’s disease. COUP-TFII overexpression in mouse dopaminergic neurons caused neurodegeneration. The goal of this study is to identify molecular mechanisms of COUP-TFII-mediated cell death through RNAseq. We utilized a previously established CAG-S-COUP-TFII allele and crossed it with the Slc6a3-Cre line to overexpress COUP-TFII specifically in dopaminergic neurons. The experimental group is Slc6a3-Cre; CAG-S-COUP-TFII, while the control group is Slc6a3-Cre. Ventral midbrain tissues from two-month-old mice were harvested for molecular profiling.
帕金森病患者体内鸡卵白蛋白上游启动子转录因子II(COUP-TFII)水平升高。在小鼠多巴胺能神经元中过表达COUP-TFII可诱导神经退行性变。本研究旨在通过RNA测序(RNAseq)解析COUP-TFII介导的细胞死亡的分子机制。我们采用已构建的CAG-S-COUP-TFII等位基因,将其与Slc6a3-Cre品系杂交,使COUP-TFII在小鼠多巴胺能神经元中特异性过表达。实验组为Slc6a3-Cre; CAG-S-COUP-TFII,对照组为Slc6a3-Cre。收集2月龄小鼠的中脑腹侧组织进行分子谱分析。
