Regulatory T (Treg) cells are involved in self tolerance and suppression of immunity to pathogens or tumours. FOXP3 is essential for Treg cell development and function. However, the FOXP3-dependent molecular mechanisms are not well understood. Here we compare the expression levels in CD4+ as well as Treg cells in reponse to different treatments in order to evaluate the Treg cell specific gene expression profile. Freshly isolated CD4+ T cells, CD4+CD25- as well as CD4+CD25+ Treg cells were treated as indicated and afterwards analysed by perfoming genome-wide expression analysis.
调节性T细胞(Regulatory T cells, Treg)参与机体自身耐受以及对病原体或肿瘤的免疫应答抑制。FOXP3对Treg细胞的发育与功能至关重要,但目前其FOXP3依赖的分子机制尚未被充分阐明。本研究通过对比不同处理条件下CD4+ T细胞与Treg细胞的基因表达水平,以期解析Treg细胞特异性基因表达谱。研究人员按照实验设计处理新鲜分离的CD4+ T细胞、CD4+CD25- T细胞以及CD4+CD25+ Treg细胞,随后采用全基因组表达分析技术完成样本检测与分析。
