To investigate the molecular mechanisms responsible for failed skeletal muscle repair in the Chronic limb threatening ischemia (CLTI) limb. We used single cell RNA sequencing (scRNA-seq) to profile the transcriptomes of the skeletal muscle specimens. Overall design: Hindlimb ischemia (HLI) or sham surgery was performed on young adult C57BL/6 and BALBc mice. Hindlimb skeletal muscle tissue was collected on post-op day 1 (sham and HLI) and post-op days 3 and 7 (HLI only). Skeletal muscle was dissociated into single cells, which were analyzed by scRNA-seq.
本研究旨在探究慢性肢体威胁性缺血(Chronic Limb Threatening Ischemia, CLTI)患肢骨骼肌修复失败的分子机制。我们采用单细胞RNA测序(Single Cell RNA Sequencing, scRNA-seq)技术对骨骼肌标本的转录组进行测序分析。实验总体设计:对年轻成年C57BL/6及BALBc小鼠实施后肢缺血(Hindlimb Ischemia, HLI)模型造模或假手术处理;分别于术后第1天(假手术组与后肢缺血组)、术后第3天和第7天(仅后肢缺血组)采集后肢骨骼肌组织。将采集的骨骼肌组织解离为单个细胞,随后通过单细胞RNA测序完成相关分析。
