As accumulating evidence suggests cancer cells exhibit stem-like properties for advanced diseases and cancer stem cells (CSCs) have been identified in various tumor types, revisiting present knowledge toward CSCs is essential. The identification of exosome-loaded miRNAs responsible for stemness and chemoradioresistance of CSCs should eventually leadd to development of novel therapeutic approaches. Overall design: Expression profiles of miRNAs of purified exosome-like vesicles (ELVs) from parental mouse CT26 cells and CT26-derived cancer spheres were generated for identifying central miRNAs for CSC malignancy.
随着日益增多的研究证据表明,晚期疾病中的癌细胞具备干细胞样特性,且癌症干细胞(Cancer Stem Cells, CSCs)已在多种肿瘤类型中被鉴定得到,重新审视当前关于癌症干细胞的研究认知至关重要。鉴定可调控癌症干细胞干性及放化疗抗性的外泌体负载微小RNAs(microRNAs, miRNAs),最终将推动新型治疗策略的开发。实验整体设计:本研究构建了源自亲本小鼠CT26细胞及CT26来源癌球的纯化外泌体样囊泡(exosome-like vesicles, ELVs)的微小RNA表达谱,用于筛选与癌症干细胞恶性表型相关的核心微小RNA。
