To elucidate the function of Nova1 in neurons, especially in inhibitory neurons, gene expression profiling analysis (RNAseq, TRAP-seq) was performed using Nova1 conditional knockout mice and their primary neurons. To analyze changes in gene expression in inhibitory neuron specific Nova1 knockout (cKO) mice, we performed RNA sequencing analysis of each brain region (hypothalamus, substantia nigra, periaqueductal gray, caudate putamen) and pituitary gland. In addition, TRAP-seq was performed on whole brains of neonatal mice to capture changes occurring in inhibitory neurons. Finally, primary neurons were used to assess the effects of Nova1 loss and the impact of overexpression of Impact, a Nova1 target gene.
