To evaluate the role of receptor geometry in interferon signaling, we engineered cell lines that express wild-type and mutant IFNλ cognate receptors (IFNλR1) with rotated intracellular registers with respect to the associated JAK1. We then performed gene expression profiling analysis using data obtained from RNA-seq of 2 different cell lines treated with 3 indicated interferons at 24hr. 为探究受体几何结构在干扰素信号转导中的作用,我们构建了可表达野生型与突变型IFNλ同源受体(IFNλR1)的基因工程化细胞系,该受体的细胞内结构域相较于与之结合的JAK1呈旋转构象。随后,我们针对经3种指定干扰素处理24小时的两种不同细胞系的RNA测序(RNA-seq)数据开展基因表达谱分析。整体实验设计:对干扰素处理后的野生型与几何结构优化型HEK293细胞的RNA测序数据进行对比基因表达谱分析。
