In this study, we investigate the anti-aging response induced by dietary restriction (DR) on gene expression level. For this, we carried out Ribosomal RNA depleted Total RNA sequencing in 16 weeks old Ercc1∆/- ad libidum (AL), DR and wt mice. Total RNA was extracted from fresh liver samples from 16 weeks old Ercc1∆/- AL, DR and wt mice. Ribosomal RNA was depleted from the extracts by using RiboMinus kit (Ambion) then sequenced according to the Illumina TruSeq v3 protocol on HiSeq2000 platform.
本研究探讨了饮食限制(dietary restriction, DR)在基因表达水平上诱导的抗衰老应答。为此,我们对16周龄的Ercc1∆/- 自由进食组(ad libitum, AL)、饮食限制组(DR)及野生型小鼠(wild type, wt)开展了核糖体RNA去除型总RNA测序。总RNA提取自上述16周龄Ercc1∆/- 自由进食组、饮食限制组及野生型小鼠的新鲜肝脏样本;随后通过Ambion公司的RiboMinus试剂盒去除提取物中的核糖体RNA,再依照Illumina TruSeq v3建库测序流程,在HiSeq2000测序平台上完成测序。
