The paralogous transcriptional cofactors Yes-associated protein (YAP) and transcriptional co-activator with PDZ-binding motif (TAZ, also called WWTR1), the main effectors of the Hippo signal transduction pathway, are emerging as pivotal determinants of malignancy in cancer. We aim to compare the transcriptome altered by knockdown of YAP, TAZ or both using siRNA in MDA-MB-231 (derived from human metastatic breast adenocarcinoma). Overall design: Knockdown of YAP, TAZ or both using siRNA in MDA-MB-231 cell line and assess the transcriptomic profile.
作为Hippo信号转导通路(Hippo signal transduction pathway)的核心效应分子,旁系同源转录辅因子Yes相关蛋白(Yes-associated protein, YAP)与携带有PDZ结合基序的转录共激活因子(transcriptional co-activator with PDZ-binding motif, TAZ,又称WWTR1)正逐渐被证实为癌症恶性表型的关键决定因素。本研究旨在以源自人转移性乳腺腺癌的MDA-MB-231细胞系为模型,采用小干扰RNA(siRNA)分别或同时敲低YAP、TAZ的表达,以此比较不同敲低条件下细胞转录组的变化差异。实验整体设计:在MDA-MB-231细胞系中利用siRNA敲低YAP、TAZ或二者联合敲低,随后检测细胞的转录组谱。
