RNA-seq was performed to reveal differences in coral responses to UV filter and heat stress. Fragments of adult stony coral Acropora tenuis were exposed to four concentrations of benzophenone-3 (BP-3) ranged from 0.38 to 2.7 mg/L or heat stress (31°C) for 96 h. RNA was extracted from each fragment (n = 6) and RNA-seq was performed on the DNBSEQ-G400 platform (2 � 200-bp).

本研究采用RNA测序(RNA-seq)技术,旨在解析珊瑚对紫外线过滤剂与热胁迫的响应差异。将成年石珊瑚纤细鹿角珊瑚(Acropora tenuis)的组织片段,分别暴露于四种浓度的二苯甲酮-3(BP-3)环境中(浓度范围为0.38~2.7 mg/L),或置于31℃的热胁迫条件下,处理时长均为96小时。从每个珊瑚组织片段(每组含6个生物学重复样本)中提取总RNA,随后在DNBSEQ-G400测序平台上开展RNA测序,测序策略为2×200 bp双端测序。
