We used single cell multi-omics method to simultaneously profiled both DNA methylome and mRNA transcriptome from single DRG sensory neurons under either control or peripheral nerve injury condition. Overall design: In this study, we performed sciatic nerve injury surgery on C57BL/6 and CAST/Ei crossing mice at different ages, and performed scMT-seq, µRNA-seq (low-input RNA-seq) and µWGBS (low-input WGBS) on sensory neurons under intact and injured condition. Bioinformatic analysis revealed novel role for DNA methylation in sensory neuron injury responses.

本研究采用单细胞多组学(single cell multi-omics)技术,对对照条件及周围神经损伤(peripheral nerve injury)条件下的单个背根神经节(Dorsal Root Ganglion, DRG)感觉神经元的DNA甲基化组(DNA methylome)与mRNA转录组(mRNA transcriptome)完成了同步分析。实验整体设计:本研究对不同周龄的C57BL/6与CAST/Ei杂交小鼠实施坐骨神经损伤手术,并分别对完整及损伤状态下的感觉神经元开展scMT-seq、µRNA-seq(低起始量RNA测序,low-input RNA-seq)与µWGBS(低起始量全基因组亚硫酸氢盐测序,low-input WGBS)。生物信息学分析揭示了DNA甲基化在感觉神经元损伤应答中的全新作用。
