Purpose: Next-generation sequencing (NGS) has provided an opportunity to understand the underline processes invloved in migratory behavior. The goals of this study are to compare NGS-derived liver transcriptome profiling (RNA-seq) under migratory and non-migratory processes and understand the processes contributign under the physiological state. Overall design: In total, 12 liver samples (6 x 2; one sample from each of the six time-points of both M and nM states) were sequenced and mapped against zebra finch genome. A range of 20-30 million 100 base single-end reads were generated for each liver sample
研究目的:下一代测序(Next-generation sequencing, NGS)为解析鸟类迁徙行为背后的潜在调控过程提供了全新契机。本研究旨在对比迁徙与非迁徙状态下,基于NGS的肝脏转录组谱(RNA测序,RNA-seq)数据差异,进而阐明该生理状态下的核心分子调控机制。
整体实验设计:本研究共纳入12份肝脏样本(6×2组,即迁徙组(M)与非迁徙组(nM)各设置6个时间点,每组每个时间点各获取1份样本),将样本进行测序后比对至斑胸草雀(Zebra finch)参考基因组。每份肝脏样本均生成2000万至3000万条100碱基单端测序读段。
