Transcriptome sequencing with short read technology usually involves either de novo assembly or mapping of the reads to a reference sequence. Both of these approaches can result in bioinformatics artefacts that lead to the output of fragmented transcripts. To overcome this problem we chose a Pacific BioSciences IsoSeq approach which can theoretically yield full-length transcripts, without the need for assembly or read mapping. We applied this approach in barley (Hordeum vulgare), using the reference cultivar 'Morex'.
采用短读长测序技术的转录组测序,通常要么开展从头组装(de novo assembly),要么将测序读段(reads)比对至参考序列。这两种方法均可能产生生物信息学伪影(bioinformatics artefacts),进而导致片段化转录本的输出。为解决该问题,本研究选用太平洋生物科学公司(Pacific BioSciences)的IsoSeq技术,该技术理论上可获得全长转录本,无需进行组装或读段比对。本研究以大麦(Hordeum vulgare)的参考栽培品种‘Morex’为实验材料,应用了该技术。
