This study investigates the diversity of archaea in a wide range of animals, to identify the dominant and rare archaeal lineages in the gut, as well as the factors influencing the structure and abundance of the archaeal community. This study is based on 341 samples collected from 250 species of animals belonging invertebrates, fish, amphibians, birds, reptiles and mammals. The 16S rRNA gene of Archaea was amplified using primers specific to this domain and sequenced with Miseq. The 16S rRNA gene of all prokaryotes was also sequenced (Miseq) after amplification with prokaryotic universal primers.

本研究针对多种动物体内的古菌(Archaea)多样性展开调查,旨在明确其肠道内的优势古菌谱系与稀有古菌谱系,并解析影响古菌群落结构与丰度的关键因素。本研究共采集了隶属于无脊椎动物、鱼类、两栖类、鸟类、爬行类及哺乳类共250个物种的341份样本。研究采用针对古菌域设计的特异性引物扩增古菌16S核糖体RNA(16S rRNA)基因,并通过MiSeq平台完成测序;同时使用原核生物通用引物扩增所有原核生物的16S rRNA基因,同样借助MiSeq平台完成测序。
