乳酸菌是一类广泛存在于自然界的革兰氏阳性细菌,在食品发酵、医药保健等领域具有重要应用价值。随着对乳酸菌功能研究的深入,人们越来越希望能够对其进行精准的遗传改造,以满足不同领域的需求。合成生物学和基因编辑技术的迅猛发展,使得利用基因编辑工具对乳酸菌进行精确的基因改造逐渐成为研究热点。
乳酸菌基因编辑主要利用了同源重组双交换和单碱基编辑技术,面向基因组改造研究,包括:基因敲除,删除特定基因以研究其功能或优化代谢途径;基因插入,引入外源基因以赋予乳酸菌新功能,如生产特定代谢物;基因调控,通过编辑启动子或调控序列,优化基因表达水平;代谢工程,改造代谢途径以提高目标产物的产量或效率。
Lactic acid bacteria (LAB) are a class of Gram-positive bacteria widely distributed in nature, holding important application values in fields such as food fermentation, medical and health care. With the deepening of research on the functions of LAB, there is an increasing demand for precise genetic modification of these bacteria to meet the requirements of different application scenarios. The rapid development of synthetic biology and gene editing technologies has gradually made precise genetic modification of LAB via gene editing tools a research hotspot.
Gene editing of LAB mainly utilizes homologous recombination double crossover and single-base editing technologies for genome modification research, which includes:
- Gene knockout: deleting specific genes to study their functions or optimize metabolic pathways;
- Gene insertion: introducing exogenous genes to endow LAB with new functions, such as producing specific metabolites;
- Gene regulation: optimizing gene expression levels by editing promoters or regulatory sequences;
- Metabolic engineering: modifying metabolic pathways to improve the yield or efficiency of target products.
