To investigate cellular population heterogeneity, and gene expression in osteoprogenitors and other cell types (endothelial, osteogenic, immune) we isolate the calvarial periosteum and sagittal sutures as stem cell compartments that contribute to osteogenesis. We compare cell populations and gene expression between different ages of mouse, and also between different diets (ad libitum vs intermittent fasting) Gene expression analysis of single cell RNA-seq data for cells isolated from the calvarial periosteum and sagittal sutures of young, adult, and aged mice, and those undergoing intermittent fasting
为探究细胞群体异质性,以及骨祖细胞(osteoprogenitors)、内皮细胞(endothelial)、成骨细胞(osteogenic)与免疫细胞(immune)等细胞类型的基因表达特征,我们将颅盖骨骨膜(calvarial periosteum)与矢状缝(sagittal sutures)这两类作为成骨过程中干细胞区室的组织进行分离。我们对比了不同月龄小鼠的细胞群体与基因表达差异,同时也比较了不同饮食干预模式下的相关指标,具体包括自由进食(ad libitum)与间歇性禁食(intermittent fasting)两组。本数据集包含对幼年、成年、老年小鼠颅盖骨骨膜与矢状缝分离细胞的单细胞RNA测序(single cell RNA-seq)数据开展的基因表达分析,同时涵盖接受间歇性禁食处理的小鼠样本的相关分析结果。
