HSC subtypes with distintic lineage restriction have been previously reported. Here we examine the transcriptome profiles of HSC subtypes with P-restricted, PEM-restricted or multilineage in vivo output. Single cell transplantation of Vwf+ HSCs from mouse bone marrow was performed to generated primary recipients showing lineage-restricted reconstitution over 16 weeks: P-restricted, PEM-restricted and multilineage. Donor-derived Gata1- HSCs were isolated from recipients' bone marrow for bulk RNA-seq (100 cells per replicate, 3 replicates for PLT-HSC, 6 replicates for PEM-HSC, 4 replicates for MUL-HSC).
