Identification and counts of specimens were performed at the inverted microscope after sedimentation of variable volumes of seawater (1-100 ml), depending on cell concentration (Utermöhl, 1958), on two transepts representing ca. 1/30 of the whole bottom area of the sedimentation chamber at 400x magnification. Specimens were identified at the species level when possible, or at higher taxonomic levels. Taxonomic identification was performed according to the most relevant and updated taxonomic literature. For selected species, the identification was checked at the electron microscope.
标本的鉴定与计数操作在倒置显微镜下进行。根据细胞浓度,取1-100 mL不等体积的海水进行沉降(Utermöhl, 1958),随后在400倍放大倍数下,对占沉降室底部总面积约1/30的两条横断区域开展观测计数。尽可能对标本进行种级鉴定,若无法实现则鉴定至更高分类阶元。分类鉴定依据最相关且最新的分类学文献开展;对选定物种,其鉴定结果通过电子显微镜进行验证。
