Extraction of DNA was performed using the CTAB method. Following the principle of uniform distribution on the chromosome, 565 simple sequence repeat (SSR) markers covering the whole genome of wheat were selected based on the high-density microsatellite consensus map. The selected SSR markers were screened for polymorphisms between the two parents, which yielded 187 polymorphic markers. The polymorphic SSR markers were then used to perform a genome-wide screen of the IL population<b>. </b>Marker type: 2 for Shaanhan8675; 0 for Lumai14; -1 for missing markers.
本研究采用CTAB(十六烷基三甲基溴化铵)法完成DNA提取。依据染色体均匀分布原则,基于高密度微卫星整合图谱,筛选得到覆盖小麦全基因组的565个简单序列重复(SSR)标记。针对两个亲本间的多态性对上述SSR标记进行筛选,最终获得187个多态性SSR标记。随后利用该批多态性SSR标记对导入系(Introgression Line, IL)群体开展全基因组筛选。标记分型规则如下:陕旱867记为2,鲁麦14记为0,标记缺失则记为-1。
