This project uses Agrobacterium tumefaciens to create a library of randomly barcoded, randomly integrated insertion mutants in Rhodosporidium toruloides. In RB-TDNAseq, the random barcodes are mapped to the genomic location of their insertion and associated with the gene they disrupt (if any). The abundance of each barcode as measured in barcode sequencing (BarSeq) experiments in differing conditions is used to infer gene function in those conditions.
