将去卵巢小鼠分为四组,分别给予雌二醇(E2)+孕酮(Progesterone)、E2+孕酮+十四烷酰佛波醇乙酸酯(TPA)、E2+孕酮+米非司酮(RU486)处理,并设置假手术组,连续处理14天;随后通过荧光激活细胞分选术(FACS)分离腔成熟细胞、腔祖细胞与乳腺干细胞,并对各细胞谱系群体开展RNA测序(RNASeq)。相较于E2+孕酮处理组小鼠,经TPA与RU486处理的小鼠乳腺干细胞池种群数量显著降低。
