The rise of antimicrobial resistance is rapidly emerging as one of the most critical global health threats with high morbidity and mortality. The resistance against our last resort antibiotics has created an urgent demand for the development of new antibiotics or modifications of current antibiotics to regain their activity for clinic use. Many of the medically relevant antibiotics are enzymatically produced by multi-modular enzymes known as non-ribosomal peptide synthetases. By integrating multidisciplinary approaches this thesis highlights the potential of engineering these biosynthetic machineries and offers a promising route to develop novel antibiotics to overcome clinical antibiotic resistance.
